UV-Vis Spectrophotometers are crucial tools for measuring the absorbance of ultraviolet and visible light by a sample. This technique identifies and quantifies various compounds. The process starts with a light source emitting UV and visible light, which passes through a monochromator to select a specific wavelength. The selected light then travels through the sample in a cuvette. As the light passes through, the sample absorbs some of it, and the remaining light reaches a detector. The detector measures the intensity of the transmitted light and compares it to the initial intensity, determining the absorbance. This absorbance directly relates to the concentration of the absorbing species in the sample, following the Beer-Lambert law. High Performance to Meet Diverse Needs
- Low Stray Light: Stray light is at 0.5 % max. (198 nm), making accurate measurements are possible up to the vicinity of 2 Abs even in the ultraviolet region. In addition, high-concentration samples can be quantified accurately.
- High Reproducibility and Repeatability Accuracy:The figure on the right is a calibration curve for caffeine, created with absorbance at 273 nm. The calibration curve has an Abs = 0.0528 Conc. The lower limit of quantitation determined from the standard deviation is 0.0051 mg/L.

- Instrument Validation Functions Compliant with JP, USP, and EP: This instrument can not only run checks for nine JIS items, but also those stipulated in the Japanese Pharmacopoeia (JP), United States Pharmacopeia (USP), and the European Pharmacopoeia (EP). The Shimadzu BioSpec-nano is a spectrophotometer suitable for carrying out concentration checks for DNA and RNA nucleic acid samples. BioSpec-nano overview
Capable of performing quantitation and purity checking of nucleic acids, quantitation of proteins, and photometric measurements (OD values displayed and printed for specified wavelengths, up to 8 wavelengths). Simply drop 1 to 2 μL of the sample onto the measurement window and press the instrument’s Start button (or click the Start Measurement button in the software window), and all steps in the process, from setting the optical path length, measurement, up until the task of wiping off the sample from the measurement window, are all carried out automatically. Troublesome work of moving arm up and down and wiping the sample from the measurement window now unnecessary. Moreover, when using the specialized software, all it takes to perform a measurement, output a report, export data, or carry out other common tasks is to click buttons on the toolbar.
Features of the BioSpec-nano
- Quick Analysis: Just drop the sample onto the target and click the button. That’s all there is to analysis. The instrument will perform sample mounting, measurement, and wiping. No need to move the wiper arm manually, nor wipe the liquid-contact parts with a cloth. Spectra measurement is conducted and the analysis data is obtained quickly.
- 1 µL – 2 µL Nucleic Acid Quantitaion*: Analysis can be performed with 1 µL (pathlength: 0.2 mm) or 2 µL (pathlength: 0.7 mm) samples. The sample is dropped onto the target with a pipette. The optional 5 mm cell allows measurement of 5 mm path length. * If the surface tension of the sample is small, droplets might not be formed with a 1 to 2 µL sample.
- Easy-to-use Software: Basic operations can be conveniently performed by clicking icons in the software or function keys on the instrument itself. Analysis results can be converted to PDF or CSV files.

The SolidSpec-3700i/3700i DUV have three detectors which cover the range from ultraviolet to near-infrared. The sensitivity in the near-infrared region is significantly enhanced by using both InGaAs and cooled PbS detectors. Conventional spectrophotometers have used a photomultiplier tube detector for the ultraviolet and visible region and a PbS detector for the near-infrared region. However, neither detector has much sensitivity near the detector switchover, preventing high sensitivity measurement in this range.
Wide Measurement Wavelength Range (SolidSpec-3700i DUV)
The development of precise laser machining using an ultraviolet laser such as an ArF excimer laser enhances the requirement for transmittance or reflectance measurements of optical parts in the deep ultraviolet region. The SolidSpec-3700i DUV(note1) enables measurement in the range of 175 nm to 2600 nm(note2) with an integrating sphere and the range of 165 nm to 3300 nm(note3) by mounting the optional Direct Detection Unit DUV. With this additional unit, the wide range from deep ultraviolet to near-infrared is now measurable.
Large Sample Compartment Accommodates a Wide Variety of Samples.
The SolidSpec-3700i & 3700i DUV have large sample compartments which allow large samples to be measured without sample destruction. Their internal dimensions are 900W × 700D × 350H mm. A sample maximum of 700W × 560D × 40H mm can be set in the sample compartment and an entire sample area of 12 inches or 310 × 310 mm is measurable by mounting the Automatic X-Y stage (option). The vertical optical path makes it possible to perform transmission or reflectance measurements of large samples.
